Zun\u00e4chst wollen wir uns den Trennmechanismus der HILIC-Methode ansehen. Hierbei handelt es sich um eine Trennung unter Verwendung polarer station\u00e4rer Phasen. Da die station\u00e4re Phase polar ist, ist die HILIC-Methode gut geeignet zur Analyse von polaren Molek\u00fclen, die an typischen Umkehrphasen nur begrenzt oder gar nicht retardiert werden. Die mobilen Phasen sind den bei der Umkehrphasen-LC verwendeten Phasen vergleichbar. Bei der HILIC-Trennung ist der Anfangseluent jedoch eine mobile Phase mit hohem organischen Anteil, wobei auf der Oberfl\u00e4che der polaren station\u00e4ren Phase eine w\u00e4ssrige Schicht erzeugt wird.<\/p>\n\n\n\n<p>Da die polaren Analyte in der w\u00e4ssrigen Schicht l\u00f6slich sind, k\u00f6nnen sie mit der polaren station\u00e4ren Phase in Wechselwirkung treten und auf mehrfache Weise retardiert werden, z. B. durch Wasserstoffbr\u00fcckenbindung, Dipol-Dipol-Wechselwirkungen und Ionenaustausch. Aufgrund der differentiellen Verteilung zwischen der station\u00e4ren Phase, der w\u00e4ssrigen Schicht und der mobilen Phase, kann das Gemisch der polaren Verbindungen dann selektiv von der HILIC-S\u00e4ule eluiert werden. Und weil die mobile Phase einen hohen Anteil des organischen L\u00f6semittels enth\u00e4lt, ist die Nachweisempfindlichkeit deutlich erh\u00f6ht, wenn ein Massenspektrometer als Detektor verwendet wird.
