Selbst mit modernen Verfahren der Lebensmittelverarbeitung lassen sich diese Verbindungen nicht vollst\u00e4ndig entfernen, so dass strikte \u00dcberwachungsprotokolle festgelegt wurden. Eine universelle Methode zur Analyse der Mykotoxine w\u00fcrde zwar ein hocheffizientes Screening erm\u00f6glichen, die Entwicklung einer derartigen Methode ist jedoch aufgrund der Unterschiede der physikochemischen Eigenschaften der Mykotoxine, der Extraktionseffizienzen und der Matrixeffekte eine gro\u00dfe Herausforderung. Zhang et al. ver\u00f6ffentlichten eine Multi-Labor-Studie [1] mit dem Ziel, den Laboren ein analytisches Verfahren zur Verf\u00fcgung zu stellen, das zur Analyse einer Vielzahl von Mykotoxinen in unterschiedlichsten Matrices geeignet ist. Aufbauend auf diesen Arbeiten entwickelten wir die folgende LC-MS\/MS-Methode, mit der sich 12 FDA-kontrollierte Mykotoxine innerhalb des Druckbereiches herk\u00f6mmlicher HPLC-Ger\u00e4te trennen lie\u00dfen.<\/p>\n\n\n\n<p>Im vorliegenden Beispiel wurden Mykotoxine in einer Erdnusspulver-Matrix analysiert. Die Verwendung einer relativ kurzen S\u00e4ule, die Selektivit\u00e4t der station\u00e4ren Phase (Biphenyl) und die Effizienz der 2.7\u00b5m-Raptor Core-Shell-Partikel erm\u00f6glichten ausgezeichnete Trennungen in einer schnellen Analysenzeit von 5.5 Minuten (bei einer Gesamtzykluszeit von 7 Minuten). Eine koeluierende Matrixverbindung zeigte denselben st\u00e4rksten MRM \u00dcbergang wie Mykotoxin HT-2 (447.3-285.3), so dass ein weniger empfindlicher \u00dcbergang (447.3-345.3) zur quantitativen Bestimmung ausgew\u00e4hlt wurde. Zur Erh\u00f6hung der Empfindlichkeit wurde ein Ammoniumpuffer verwendet, um eine bessere Ionisierung der Mykotoxine zu f\u00f6rdern. Die Raptor Biphenyl S\u00e4ule eignete sich sehr gut f\u00fcr die 12 Mykotoxine, die in der zitierten Studie untersucht wurden. F\u00fcr erweiterte Listen von Substanzen, die isobare Mykotoxine \u00e4hnlicher Strukturen enthalten, ist m\u00f6glicherweise die Raptor FluoroPhenyl Phase erforderlich, um ausreichende chromatografische Aufl\u00f6sung zu erzielen. Die passende Selektivit\u00e4t der Raptor FluoroPhenyl-S\u00e4ule wurde in einer&nbsp;<a href=\"https:\/\/discover.restek.com\/articles\/ffss2790\/highly-selective-lc-msms-analysis-of-mycotoxins-from-multiple-classes\" data-type=\"post\" data-id=\"43565\">Analyse von 20 Mykotoxinen<\/a>&nbsp;unter Beweis gestellt.<br><br>Die vorliegende Methode zeigte ausgezeichnete Pr\u00e4zision und Richtigkeit f\u00fcr die 12 FDA-kontrollierten Mykotoxine, die im Rahmen einer Validierungsstudie mit einer Vielzahl von Matrices untersucht wurden (darunter verschiedene Quellen von Maismehl und braunem Reismehl neben dem hier gezeigten Erdnusspulver).
