Diese Produkte werden aus diversen getrockneten Pflanzen hergestellt, die aufgrund landwirtschaftlicher Praktiken Pestizidr\u00fcckst\u00e4nde enthalten k\u00f6nnen. Zur Sicherung ihrer Qualit\u00e4t und um eine Exposition zu vermeiden, ist ihre \u00dcberwachung deshalb unerl\u00e4sslich. Die Inhaltsstoffe liegen in getrocknetem Zustand vor, so dass die Standard-QuEChERS-Methoden [1] modifiziert werden m\u00fcssen. Die beste Methode zur Trennung von Mehrstoffgemischen mit koextrahierten St\u00f6rsubstanzen ist die Gaschromatografie. In Verbindung mit einem hochempfindlichen Triple-Quadrupol-Massenspektrometer (GC-MS\/MS) lassen sich so Verunreinigungen im Spurenbereich nachweisen. In dieser Studie haben wir auf einem Triple-Quadrupol GC-MS\/MS-System mit der modifizierten QuEChERS-Methode mehr als 200 Verbindungen gleichzeitig analysiert.&nbsp;<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Materialien und Methoden<\/h2>\n\n\n\n<p>Pestizidstandards, interne Standards, Qualit\u00e4tskontrollstandards und ein QuEChERS-Kit wurden von Restek erhalten:<\/p>\n\n\n\n<ul class=\"wp-block-list\">\n<li>GC Multipestizid-Kit (Art.-Nr. 32562)<\/li>\n\n\n\n<li>QuEChERS Internal Standard Mix f\u00fcr die GC-MS Analyse (Art.-Nr. 33267)<\/li>\n\n\n\n<li>SV Internal Standard Mix (Art.-Nr.31206)<\/li>\n\n\n\n<li>Q-sep QuEChERS Extraktions-Kit (Urspr\u00fcngliche Art.-Nr. 23991 wurde abgek\u00fcndigt; siehe\u00a0Art.-Nr.\u00a025848)<\/li>\n<\/ul>\n\n\n\n<p>Insgesamt wurden 232 Verbindungen (220 Pestizide, 6 interne Standards und 6 Qualit\u00e4ts-kontrollstandards) analysiert. Kommerziell erh\u00e4ltliches Ginseng wurde als Matrix verwendet. Damit wurden matrixangepasste Kalibrierstandards (1 bis 200 ng\/mL) und angereicherte Proben (jeweils zwei mit 10 ng\/g und 50 ng\/g) hergestellt. Kalibrierkurven (gewichtet 1\/C) wurden mithilfe des internen Standards PCB52 generiert.&nbsp;<\/p>\n\n\n\n<p>Eine MRM-Analysemethode wurde unter Verwendung der Smart Pesticides Database (Shimadzu) erstellt. Diese Datenbank enth\u00e4lt Retentionsindizes f\u00fcr alle registrierten Verbindungen und die Retentionszeiten lassen sich durch Analyse eines n-Alkan-Probengemischs vorhersagen (AART: Automatic Adjustment of Retention Time). Auf der Basis der erwarteten Retentionszeiten erzeugt Smart MRM ein optimales Zeitprogramm zur Datenerfassung (Abbildung 1).&nbsp;<\/p>\n\n\n<div class=\"wp-block-custom-chromatogram-article-top\"><div class=\"chromatogram-article-placeholder\"><div class=\"figure-heading\"><strong>Abbildung 1:\u00a0<\/strong>\u00a0Von Smart MRM erstelltes Zeitprogramm zur Datenerfassung<\/div><div class='chromatogram-article-inner-full'><div class=\"chromatogram-article-inner\">\n<div class=\"wp-block-image\">\n<figure class=\"aligncenter size-large\"><img loading=\"lazy\" decoding=\"async\" width=\"1024\" height=\"960\" src=\"https:\/\/discover.restek.com\/wp-content\/uploads\/\/figure-article-FSAR3080-01-1024x960.jpg\" alt=\"\" class=\"wp-image-20041\" title=\"-\" srcset=\"https:\/\/discover.restek.com\/wp-content\/uploads\/figure-article-FSAR3080-01-1024x960.jpg 1024w, https:\/\/discover.restek.com\/wp-content\/uploads\/figure-article-FSAR3080-01-300x281.jpg 300w, https:\/\/discover.restek.com\/wp-content\/uploads\/figure-article-FSAR3080-01-768x720.jpg 768w, https:\/\/discover.restek.com\/wp-content\/uploads\/figure-article-FSAR3080-01-1536x1440.jpg 1536w, https:\/\/discover.restek.com\/wp-content\/uploads\/figure-article-FSAR3080-01.jpg 1800w\" sizes=\"auto, (max-width: 1024px) 100vw, 1024px\" \/><\/figure>\n<\/div>\n<\/div><\/div><\/div>\n\n\n<div style=\"height:20px\" aria-hidden=\"true\" class=\"wp-block-spacer\"><\/div>\n\n\n\n<h2 class=\"wp-block-heading\">Extraktion und Reinigungsverfahren<\/h2>\n\n\n\n<ul class=\"wp-block-list\">\n<li>1.0 \u00b1 0.05 g gemahlenes Ginsengpulver in 50-mL-Polypropylen-Zentrifugenr\u00f6hrchen einwiegen.<\/li>\n\n\n\n<li>10 mL Wasser von HPLC-Qualit\u00e4t zugeben und das R\u00f6hrchen auf einem Vortex-Mischer kr\u00e4ftig sch\u00fctteln.<\/li>\n\n\n\n<li>10 mL des ACN\/IS-Extraktionsl\u00f6semittels zugeben.<\/li>\n\n\n\n<li>R\u00f6hrchen 15 Minuten stehen lassen.<\/li>\n\n\n\n<li>4 g wasserfreies MgSO4 und 1 g NaCl zugeben.<\/li>\n\n\n\n<li>R\u00f6hrchen auf einem mechanischen Sch\u00fcttelapparat bei hoher Sch\u00fcttelintensit\u00e4t 30 Minuten lang sch\u00fctteln.<\/li>\n\n\n\n<li>Die 50-mL-R\u00f6hrchen bei 3000\u20134500 U\/min 5 Minuten zentrifugieren.<\/li>\n\n\n\n<li>Die GCB\/PSA (0.25 g\/0.5 g) SPE-S\u00e4ulen mit ca. 250 mg oben aufgebrachtem wasserfreiem Na2SO4 und dem 3-fachen S\u00e4ulenvolumen Aceton konditionieren.<\/li>\n\n\n\n<li>Sammelgestell mit 15-mL-Einweg-Glaszentrifugenr\u00f6hrchen auf einem SPE-Vakuumverteiler einsetzen.<\/li>\n\n\n\n<li>1.25 mL des ACN-Extrakts zugeben.<\/li>\n\n\n\n<li>Mit 1 mL Aceton sp\u00fclen.<\/li>\n\n\n\n<li>Mit 12 mL eines Aceton-Toluol-Gemischs (3:1 Vol.\/Vol.) eluieren.<\/li>\n\n\n\n<li>Das Elutionsmittel vorsichtig (50 \u00b0C) bis auf ca. 100 \u03bcL verdampfen.<\/li>\n\n\n\n<li>500 \u03bcL Toluol zu den Leer-\/angereicherten Proben, den Kalibrierstandardl\u00f6sungen und den matrixangepassten Kalibrierstandards hinzuf\u00fcgen.<\/li>\n\n\n\n<li>20 \u03bcL Qualit\u00e4tskontrollstandards (12.5 \u03bcg\/mL) und ca. 50 mg wasserfreies MgSO4 zu allen Proben hinzuf\u00fcgen.<\/li>\n\n\n\n<li>5 Sekunden lang auf dem Vortex-Mischer sch\u00fctteln.<\/li>\n\n\n\n<li>R\u00f6hrchen 5 Minuten bei 3000 g zentrifugieren.<\/li>\n\n\n\n<li>Toluolextrakt mithilfe einer Pasteur-Pipette an die ALS GC-Fl\u00e4schchen \u00fcberf\u00fchren.<\/li>\n<\/ul>\n\n\n\n<h2 class=\"wp-block-heading\">Ergebnis und Diskussion<\/h2>\n\n\n\n<h3 class=\"wp-block-heading\">Matrixangepasste Kalibrierung<\/h3>\n\n\n\n<p>Das Chromatogramm in Abbildung 2 zeigt einen matrixangepassten 10-ng\/mL-Kalibrierstandard. Von den 232 Verbindungen konnten 230 innerhalb von \u00b10.1 Minuten der erwarteten Retentionszeit durch AART nachgewiesen werden. Die zwei verbleibenden Verbindungen, 1,4-Dichlorbenzol-d4 und Naphthalin-d8 aus den sechs Qualit\u00e4tskontrollstandards, eluierten in weniger als 4 Minuten. Zur Quantifizierung der 10-ng\/g-Konzentration werden Kalibrierpunkte mit 2 ng\/g oder weniger ben\u00f6tigt. Wie oben erw\u00e4hnt wurden die angereicherten Proben auf 2.5 ng\/mL und 12.5 ng\/mL verd\u00fcnnt. Da die Kalibrierkurven gute Linearit\u00e4t bei niedrigen Konzentrationen zeigten und die modifizierte QuEChERS-Methode Interferenzen unterdr\u00fcckte, lie\u00dfen sich gute Wiederfindungsergebnisse erzielen. (Einige Verbindungen lie\u00dfen sich nicht korrekt quantifizieren, da sie in der Matrix f\u00fcr die Kalibrierstandards urspr\u00fcnglich enthalten waren. Die Ionenverh\u00e4ltnisse zwischen dem Zielanalyt und der Referenz wurden mit dem des 100-ng\/g-Standards verglichen und gem\u00e4\u00df SANCO\/12571\/2013 [2] ausgewertet. Danach wird die Verwendung des relativen Verh\u00e4ltnisses mit einem Toleranzbereich von \u00b130 % als Kriterium empfohlen.<\/p>\n\n\n\n<p>Tabelle I zeigt die Wiederfindung und das relative Ionenverh\u00e4ltnis f\u00fcr alle Verbindungen und Abbildung 5 zeigt das Kombinationsdiagramm der Wiederfindung und des relativen Ionenverh\u00e4ltnisses, das aus dieser Tabelle generiert wurde. 76 % der Verbindungen in der Spalte 10 ng\/g-1 lagen innerhalb von \u00b130 % des relativen Ionenverh\u00e4ltnisses mit guter Wiederfindung zwischen 70 und 120 %. diese Gruppe bestand in erster Linie aus Verbindungen mit einem niedrigen Siedepunkt. Sie sind m\u00f6glicherweise bei der Verdampfung verloren gegangen. Gruppe B zeigte hohe relative Ionenverh\u00e4ltnisse, verursacht durch Matrixinterferenzen. Gruppe C zeigte eine hohe Wiederfindung; diese Gruppe bestand aus angereicherten Proben von 10 ng\/g. Einige dieser Verbindungen lagen urspr\u00fcnglich in der Matrix vor und wurden falsch quantifiziert. Bei anderen wurden die hohen Werte durch ihre \u00dcberg\u00e4nge verursacht, die niedrige Response und geringe Stabilit\u00e4t hatten. Gruppe D zeigte ein niedriges relatives Ionenverh\u00e4ltnis. Hier mussten \u00dcberg\u00e4nge mit h\u00f6herer Response festgelegt werden. Durch \u00c4nderung einiger Verfahren und Parameter lassen sich die Positionen dieser Verbindungen m\u00f6glicherweise verbessern.&nbsp;<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Schlussfolgerung<\/h2>\n\n\n\n<p>Diese Studie zeigt, dass die modifizierte QuEChERS-Methode in Verbindung mit GC-MS\/MS zur konsistenten \u00dcberwachung von Pestiziden in pflanzlichen Inhaltsstoffen geeignet ist.<\/p>\n\n\n\n<p>Obwohl getrocknete Proben oft mit einer schweren und schwierigen Matrix verbunden sind, lie\u00dfen sich mithilfe der modifizierten QuEChERS-Methode, einer SPE-S\u00e4ulenreinigung und der Verd\u00fcnnung mit Toluol matrixbedingte Interferenzen unterdr\u00fccken. Mittels GC-MS\/MS lie\u00dfen sich selbst bei verd\u00fcnnten Proben \u00e4u\u00dferst kleine Pestizidmengen nachweisen. Die Analysemethode dauert insgesamt nur 30 Minuten und deckt mehr als 200 Pestizide ab.
