In order for this analysis to be most effective, we need to prepare our samples carefully, including a filtration step, and then analyze via liquid chromatography with a UV detector. We\u2019ll walk through the steps of this important analysis with some commercially available CBD oils.&nbsp;<\/p>\n\n\n\n<h3 class=\"wp-block-heading\">Additional Resources<\/h3>\n\n\n\n<ul class=\"wp-block-list\">\n<li><a href=\"https:\/\/discover.restek.com\/articles\/FFSS2851\/high-throughput-analysis-of-cannabinoids-by-lc-uv\/\">Featured Application &#8211; Cannabinoid Profiles on Raptor ARC-18<\/a><\/li>\n<\/ul>\n\n\n\n<h3 class=\"wp-block-heading\">Restek Recommended Tools &amp; Parts<\/h3>\n\n\n\n<ul class=\"wp-block-list\">\n<li><a href=\"https:\/\/www.restek.com\/c\/1787\" target=\"_blank\" rel=\"noreferrer noopener\">Raptor ARC-18 LC Columns<\/a><\/li>\n\n\n\n<li><a href=\"https:\/\/www.restek.com\/c\/1337\" target=\"_blank\" rel=\"noreferrer noopener\">Thomson SINGLE StEP Filter Vials<\/a><\/li>\n<\/ul>\n\n\n\n<h2 class=\"wp-block-heading\">Transcript<\/h2>\n\n\n\n<p>Thanks for joining us for\u202fthis video where we\u2019ll discuss a critical workflow for cannabis analysis.&nbsp;We\u2019re going to\u202fdemonstrate the\u202fHigh-Throughput Analysis of Cannabinoids by LC-UV.&nbsp;We\u2019re specifically going to be looking at cannabinoids in CBD oils.\u202fFor this workflow, you will need the following items:&nbsp;&nbsp;<\/p>\n\n\n\n<p>Thompson SINGLE&nbsp;StEP&nbsp;vials, a Raptor ARC-18 analytical column, the matching guard column and an EXP direct connect holder.&nbsp;<\/p>\n\n\n\n<p>Our first step is to\u202fpipette\u202f50\u202f\u00b5l\u202fof\u202fCBD oil,\u202fand&nbsp;then\u202fextract\u202fthe sample\u202fwith 950\u202f\u00b5l\u202fof&nbsp;methanol,&nbsp;followed by\u202fvortexing\u202ffor 30 seconds&nbsp;at 3000 rpm.&nbsp;Next,&nbsp;a&nbsp;750\u202f\u00b5l\u202fsample solution is placed into a new amber vial.&nbsp;We then add\u202f250\u202f\u00b5l\u202fof water&nbsp;into this&nbsp;in order to&nbsp;initiate matrix precipitation.\u202f&nbsp;<\/p>\n\n\n\n<p>Now, vortex the new sample solution again, this time&nbsp;for 10 seconds&nbsp;at 3000 rpm.\u202fNext, we need to filter our sample. This is traditionally done\u202fusing syringe filters;&nbsp;however, this is a&nbsp;labor-intensive&nbsp;sample prep technique\u202fespecially\u202fwhen preparing multiple samples.&nbsp;We can prepare samples faster and generate less waste by using\u202fThomson SINGLE\u202fStEP\u202ffilter\u202fvials.\u202f&nbsp;<\/p>\n\n\n\n<p>Place\u202f400\u202ful\u202fof\u202fthe\u202fsample\u202fextract\u202finto a Thomson SINGLE\u202fStEP\u202ffilter vial.&nbsp;Then, place the plunger inside the filter vial and depress it to filter the sample extract.&nbsp;This allows the sample\u202fextract\u202fto pass through the membrane,&nbsp;filtering out particulates.\u202f&nbsp;<\/p>\n\n\n\n<p>Once&nbsp;all of&nbsp;your samples are prepped, place them in the&nbsp;autosampler&nbsp;tray.\u202fBefore you start your analysis,&nbsp;make sure your mobile phases are freshly prepared and\u202fthat you have sufficient mobile phase for the number of samples&nbsp;you are analyzing.\u202fIn addition, using Bluestem filters helps provide clean mobile phase to extend the lifetime of your column and pump seals.\u202fFor this method,\u202fwe use a\u202fRaptor ARC-18 analytical column and guard cartridge.&nbsp;Place a Raptor ARC-18 EXP guard&nbsp;cartridge into the&nbsp;direct connect holder.&nbsp;<\/p>\n\n\n\n<p>Once you\u2019ve established the method according to your lab\u2019s validation SOPs,\u202fnow&nbsp;you\u2019re&nbsp;ready to run&nbsp;your&nbsp;samples and collect data.\u202fAll compounds\u202fare resolved in this\u202ffast 9-minute analysis, making\u202fthe\u202fmethod suitable for high-throughput cannabis testing labs.&nbsp;In addition, this analysis uses a simple isocratic mobile&nbsp;phase&nbsp;that&#8217;s&nbsp;easily transferable between instruments,&nbsp;compared to more complex methods that incorporate atypical mobile phase gradients or additives.\u202f&nbsp;<\/p>\n\n\n\n<p>Thank you for joining us as we demonstrated this workflow solution for the analysis of cannabinoids in CBD oil.\u202fYou can\u202ffind&nbsp;all of&nbsp;the necessary details\u202fof both the\u202ffeatured application\u202fand my&nbsp;poster containing information&nbsp;regarding the analysis of cannabinoids in CBD oils&nbsp;in the links below or at&nbsp;www.restek.com<\/p>\n\n\n\n<div style=\"height:100px\" aria-hidden=\"true\" class=\"wp-block-spacer\"><\/div>\n","protected":false},"excerpt":{"rendered":"<p>Curious about cannabis potency analysis in CBD oil?
