De par ces conditions initiales fortement organiques, il est donc important de bien conditionner votre syst\u00e8me et votre colonne afin d\u2019obtenir les meilleures performances chromatographiques. class=\"wp-block-heading\">Transcription<\/h2>\n\n\n\n<p>Vous avez une nouvelle colonne HILIC et souhaitez d\u00e9velopper une m\u00e9thode d\u2019analyse avec ce mode.&nbsp;<br>Par quoi commencer ?&nbsp;&nbsp;<br>Comme nous en avons discut\u00e9 au pr\u00e9alable, pour les s\u00e9parations HILIC,&nbsp;on utilise des conditions initiales \u00e0 fort pourcentage d\u2019organique&nbsp;pour cr\u00e9er une couche riche en eau \u00e0 la surface de la phase stationnaire polaire.&nbsp;Il est donc n\u00e9cessaire de conditionner puis d\u2019\u00e9quilibrer la colonne entre chaque analyse&nbsp;pour r\u00e9tablir la couche aqueuse \u00e0 la surface des particules.&nbsp;<\/p>\n\n\n\n<p>Quand vous commencez \u00e0 utiliser une nouvelle colonne HILIC ou quand vous changez la composition des phases mobiles,&nbsp;nous vous recommandons de rincer la colonne avec au moins 50 volumes de colonne de phase mobile.&nbsp;De plus, au moins 10 injections de blanc,&nbsp;dans les conditions d\u2019analyse, doivent \u00eatre effectu\u00e9es lorsque vous d\u00e9marrez une nouvelle s\u00e9quence d\u2019analyses.&nbsp;Le temps d\u2019\u00e9quilibration entre chaque injection&nbsp;est tr\u00e8s d\u00e9pendant de l\u2019analyte, donc assurez-vous de bien observer la reproductibilit\u00e9 des temps de r\u00e9tention&nbsp;pour d\u00e9terminer le temps n\u00e9cessaire \u00e0 la&nbsp;r\u00e9-\u00e9quilibration.&nbsp;<\/p>\n\n\n\n<p>Maintenant, en plus de la colonne,&nbsp;nous tenons \u00e0 vous rappeler que&nbsp;votre syst\u00e8me LC doit \u00e9galement \u00eatre conditionn\u00e9 pour les analyses en mode HILIC.&nbsp;Nous savons qu\u2019une grande majorit\u00e9 des syst\u00e8mes LC sont utilis\u00e9s en phase inverse.&nbsp;Donc avant de passer \u00e0 des analyses HILIC,&nbsp;assurez-vous de rincer le syst\u00e8me avec une phase mobile fortement organique&nbsp;sur une p\u00e9riode de 30 \u00e0 60
