La plupart du temps, la colonne que vous recevez n\u2019est pas imm\u00e9diatement pr\u00eate \u00e0 l\u2019emploi, \u00e0 moins que la phase mobile que vous allez utiliser ne soit strictement identique au solvant de stockage de la colonne. La nature du solvant de stockage est sp\u00e9cifi\u00e9e sur le certificat d\u2019analyse, que vous pourrez consulter ou t\u00e9l\u00e9charger \u00e0 partir du lien internet fourni \u00e0 l\u2019int\u00e9rieur de la bo\u00eete de la colonne. La plupart de nos colonnes sont stock\u00e9es dans des m\u00e9langes de solvants contenant au moins 50% de solvant organique.&nbsp;Si vous utilisez une phase mobile diff\u00e9rente du solvant de stockage, il convient d\u2019abord de \u201crincer\u201c la colonne avec une phase mobile \u00e0 la composition en solvant organique 10% ou 20% plus faible. Faites passer un volume \u00e9quivalent \u00e0 10 volumes de colonne avant de basculer ensuite sur la phase mobile que vous allez utiliser dans votre m\u00e9thode. Pour calculer le volume de la colonne, vous pouvez utiliser l\u2019une de ces 2 formules. Notez que la valeur de la constante est diff\u00e9rente selon que vous utilisez une particule de silice compl\u00e9tement poreuse ou superficiellement poreuse.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Si vous utilisez des tampons dans votre m\u00e9thode, vous devrez d\u2019abord \u201crincer\u201c la colonne avec une phase mobile \u00e0 la composition en solvant organique 10 \u00e0 20% plus faible que celle du solvant de stockage, ceci afin de vous assurer qu\u2019il n\u2019y ait pas de pr\u00e9cipitation des sels lorsque vous introduirez le tampon, car cela pourrait conduire \u00e0 une augmentation de la contre-pression et au final boucher la colonne. Apr\u00e8s avoir fait passer vos 10 volumes de colonne, basculez sur la phase mobile tamponn\u00e9e que vous allez utiliser, et \u201crincer\u201c \u00e0 nouveau la colonne avec 10 volumes de colonne.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Surveillez la ligne de base et attendez qu\u2019elle se stabilise en termes de pression car cela vous indiquera que la colonne est \u00e9quilibr\u00e9e. De nombreux utilisateurs aiment faire une ou deux injections de conditionnement avant de commencer leurs analyses. C\u2019est un bon moyen pour pr\u00e9parer la colonne \u00e0 \u00eatre utilis\u00e9e dans les conditions pr\u00e9vues pour votre m\u00e9thode.
