Int\u00e9ressons-nous aux interactions secondaires avec les silanols, et aux moyens de les att\u00e9nuer pendant la phase de d\u00e9veloppement. Lorsque je commence \u00e0 examiner les train\u00e9es de pics et \u00e0 chercher ce qui pourrait aider \u00e0 les r\u00e9duire, je commence par observer toutes les conditions analytiques que j\u2019ai mises en place dans ma m\u00e9thode. &nbsp;<\/p>\n\n\n\n<p>Une fois pass\u00e9es en revue les phases mobiles que j\u2019ai choisies, le gradient, les temp\u00e9ratures, etc., je m\u2019attarde un peu plus sur la structure des analytes. Si je regarde les analytes, la charge positive des compos\u00e9s, concr\u00e8tement, tout ce qui peut contenir une fonction amine, comme un m\u00e9dicament faiblement basique, peut interagir avec la charge n\u00e9gative des silanols r\u00e9siduels pr\u00e9sents \u00e0 la surface de la silice. &nbsp;<\/p>\n\n\n\n<p>C\u2019est cette interaction qui cr\u00e9e les train\u00e9es de pics. Une fa\u00e7on simple de contourner ce probl\u00e8me est d\u2019ajouter un tampon dans vos phases mobiles. Lors des analyses LC-MS habituelles, nous ajoutons d\u00e9j\u00e0 un acide aux \u00e9luants aqueux et organiques de nos phases mobiles. L\u2019ajout de sels compl\u00e9mentaires, comme le formiate d\u2019ammonium dans l\u2019acide formique, peut vraiment aider \u00e0 r\u00e9duire certaines de ces interactions avec les silanols. &nbsp;<\/p>\n\n\n\n<p>En effet, la charge positive du sel tampon que nous ajoutons peut interagir avec la charge n\u00e9gative des silanols \u00e0 la surface de la silice et cr\u00e9er cette interaction. Elle va emp\u00eacher votre analyte d\u2019interagir avec la surface de la silice. Le m\u00e9canisme de r\u00e9tention primaire est provoqu\u00e9 par la phase stationnaire que vous avez choisie.
