Mais \u00e0 quoi faire attention pour obtenir les meilleurs r\u00e9sultats ?<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Dans ce \u201cConseil Restek\u201d, nous allons vous montrer comment optimiser vos injections \u201csplit\u201d.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Et nous allons commencer par les inserts d\u2019injection. Les d\u00e9bits plus \u00e9lev\u00e9s li\u00e9s \u00e0 l\u2019injection \u201csplit\u201d peuvent avoir de nombreux avantages, mais en utilisant le mauvais insert d\u2019injection, ces d\u00e9bits \u00e9lev\u00e9s peuvent \u00eatre \u00e0 l\u2019origine de nombreux probl\u00e8mes.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Pour les analyses \u201csplit\u201d, il vous faut un insert qui permette de vaporiser les analytes enti\u00e8rement et de les transf\u00e9rer dans la colonne pendant la br\u00e8ve p\u00e9riode de leur s\u00e9jour dans l\u2019injecteur. Un insert avec une obstruction comme de la laine pr\u00e9sente une plus grande surface sp\u00e9cifique qui aide \u00e0 la vaporisation. La laine rend aussi l\u2019\u00e9chantillon plus homog\u00e8ne et aide \u00e0 retenir les mati\u00e8res non volatiles, ce qui prolonge la dur\u00e9e de vie de la colonne. Les inserts ouverts en bas, comme les inserts \u201cdroits\u201d avec de la laine ou les inserts \u201cPrecision\u201d avec de la laine sont recommand\u00e9s puisqu\u2019ils ils permettent un \u00e9coulement libre vers la sortie \u201csplit\u201d.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Le \u201csplit ratio\u201d est essentiel dans l\u2019injection \u201csplit\u201d car il d\u00e9termine la quantit\u00e9 d\u2019\u00e9chantillon qui passe dans la sortie \u201csplit\u201d par rapport \u00e0 celle qui passe dans la colonne. Pour les \u00e9chantillons tr\u00e8s concentr\u00e9s, un \u201csplit ratio\u201d \u00e9lev\u00e9 peut \u00eatre n\u00e9cessaire pour \u00e9viter de surcharger la &nbsp;colonne. Si l\u2019\u00e9chantillon n\u2019est pas tellement concentr\u00e9, un \u201csplit ratio\u201d moins \u00e9lev\u00e9 vous aidera \u00e0 atteindre les limites de d\u00e9tection exig\u00e9es. Ensuite, il faut optimiser la temp\u00e9rature de l\u2019injecteur. La temp\u00e9rature doit \u00eatre assez \u00e9lev\u00e9e pour atteindre une vaporisation suffisante des compos\u00e9s les plus lourds mais suffisamment basse pour \u00e9viter une activit\u00e9 excessive pour les analytes r\u00e9actifs. 250 degr\u00e9s Celsius sont un bon point de d\u00e9part mais il peut \u00eatre n\u00e9cessaire d\u2019ajuster cette temp\u00e9rature pour obtenir les meilleurs pics sur l\u2019ensemble de l\u2019analyse.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Qu\u2019en est-il de la temp\u00e9rature initiale du four ? Du fait du transfert rapide des compos\u00e9s dans la colonne avec les injections \u201csplit\u201d, l\u2019effet de solvant est g\u00e9n\u00e9ralement inutile. C\u2019est pour cette raison qu\u2019il est souvent possible de commencer avec une temp\u00e9rature de four plus \u00e9lev\u00e9e. Mais si les analytes sont tr\u00e8s volatils, une temp\u00e9rature initiale plus faible sera n\u00e9cessaire pour une meilleure r\u00e9tention et un effet de solvant efficaces. Il n\u2019est g\u00e9n\u00e9ralement pas n\u00e9cessaire de maintenir la temp\u00e9rature initiale du four pour les injections \u201csplit\u201d, car le transfert des analytes dans la colonne est rapide.
