Les tampons sont tr\u00e8s importants en mode HILIC car les \u00e9tats d\u2019ionisation des analytes ET des phases stationnaires vont impacter la s\u00e9paration. Une bonne s\u00e9paration en mode HILIC d\u00e9pend des \u00e9tats d\u2019ionisation de la phase stationnaire ET des&nbsp;analytes, qui sont contr\u00f4l\u00e9s par le pH des phases mobiles.&nbsp;<\/p>\n\n\n\n<p>Pour commencer, nous recommandons de pr\u00e9parer au moins 3 phases mobiles aqueuses \u00e0 diff\u00e9rents pH. La premi\u00e8re est du formiate d\u2019ammonium 10mM ajust\u00e9e \u00e0 pH 3 avec une solution d\u2019acide formique. La seconde est de l\u2019ac\u00e9tate d\u2019ammonium 10mM ajust\u00e9e \u00e0 pH 5 avec une solution d\u2019acide ac\u00e9tique, et la troisi\u00e8me est de l\u2019ac\u00e9tate d\u2019ammonium 10mM ajust\u00e9e \u00e0 pH 6.8 avec une solution d\u2019acide ac\u00e9tique.&nbsp;<\/p>\n\n\n\n<p>En travaillant en&nbsp;isocratique&nbsp;ou en gradient avec des conditions initiales \u00e0 90 \u2013 95% d\u2019ac\u00e9tonitrile,&nbsp;vous devriez savoir quel pH est le plus appropri\u00e9 simplement en regardant les formes\/sym\u00e9tries des pics. Une fois le pH de la phase mobile d\u00e9termin\u00e9, vous pouvez ensuite optimiser la m\u00e9thode en ajustant la concentration du tampon et des conditions d\u2019\u00e9lution. Parfois, vous constaterez qu\u2019une concentration de tampon plus \u00e9lev\u00e9e peut donner de meilleures formes\/sym\u00e9tries de pics. Mais gardez bien en t\u00eate que le sel contenu dans le tampon peut pr\u00e9cipiter en pr\u00e9sence d\u2019une forte teneur en solvant organique. Enfin, pour les \u00e9lutions de gradient, une bonne pratique consiste \u00e0 tamponner la phase mobile aqueuse ET la phase mobile organique pour garder une force ionique constante durant le temps de l\u2019analyse.
