La d\u00e9rivation pr\u00e9-colonne, suivie d\u2019une d\u2019analyse LC en phase inverse, est une approche typique, cette d\u00e9rivation \u00e9tant n\u00e9cessaire en raison d\u2019une faible r\u00e9tention chromatographique et d\u2019une mauvaise sensibilit\u00e9 pour les acides amin\u00e9s libres. Mais ces m\u00e9thodes sont souvent chronophages et assez laborieuses. L\u2019utilisation d\u2019acides perfluor\u00e9s comme r\u00e9actifs d\u2019appariement d\u2019ions, afin d\u2019am\u00e9liorer la r\u00e9tention des acides amin\u00e9s non-d\u00e9riv\u00e9s sur une colonne C18, est une autre technique courante, mais ce genre d\u2019analyses peut avoir un impact n\u00e9gatif sur le syst\u00e8me chromatographique et le spectrom\u00e8tre de masse.<\/p>\n\n\n\n<p>L\u2019approche pr\u00e9sent\u00e9e ici est plus simple, car les acides amin\u00e9s non-d\u00e9riv\u00e9s peuvent \u00eatre analys\u00e9s directement en LC-MS\/MS \u2013 suivant une pr\u00e9paration d\u2019\u00e9chantillons simple en une seule \u00e9tape \u2013 en utilisant une colonne Raptor Polar X. Les colonnes Raptor Polar X sont faites d\u2019une phase hybride (HILIC et \u00c9change Ionique) utilisant diff\u00e9rents m\u00e9canismes d\u2019interactions qui fournissent la r\u00e9tention et la r\u00e9solution n\u00e9cessaires \u00e0 l\u2019analyse simultan\u00e9e d\u2019une grande vari\u00e9t\u00e9 de compos\u00e9s aux chimies et structures diff\u00e9rentes. Comme d\u00e9montr\u00e9 ici, des acides amin\u00e9s non-d\u00e9riv\u00e9s aux cha\u00eenes lat\u00e9rales polaires, non polaires, charg\u00e9es positivement et charg\u00e9es n\u00e9gativement, ont \u00e9t\u00e9 correctement retenus et s\u00e9par\u00e9s avec<br>un gradient court de 10 minutes. Ce dernier a \u00e9galement permis de s\u00e9parer les contaminants du syst\u00e8me des analytes d\u2019int\u00e9r\u00eat, augmentant la robustesse de la m\u00e9thode. En suivant cette m\u00e9thode, les proc\u00e9dures de pr\u00e9paration d\u2019\u00e9chantillons chronophages et laborieuses, qui peuvent inclure de co\u00fbteux kits de d\u00e9rivation, sont remplac\u00e9es par une simple pr\u00e9cipitation de prot\u00e9ines puis une analyse directe de l\u2019extrait. L\u2019analyse directe d\u2019acides amin\u00e9s non-d\u00e9riv\u00e9s sur une colonne Raptor Polar X fournit d\u2019excellents r\u00e9sultats, tout en restant simple et rapide, ce qui en fait une
