Nel presente studio \u00e8 stato sviluppato un flusso di lavoro efficace per eseguire questa analisi complessa sui brownie, e vengono illustrate in dettaglio alcune strategie di ottimizzazione volte a fornire un punto di partenza per matrici simili. Sono stati impiegati i metodi LC-MS\/MS e GC-MS\/MS e per tutti i composti target sono stati raggiunti eccellenti risultati a livello di LOQ, linearit\u00e0, accuratezza e precisione.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Introduzione<\/h2>\n\n\n\n<p>La legalizzazione della cannabis (marijuana) per scopi ricreativi e medici richiede metodi analitici accurati e affidabili per valutare la qualit\u00e0 e la sicurezza di qualsiasi prodotto derivato dalla cannabis. Per sviluppare metodi efficaci occorre considerare attentamente le propriet\u00e0 degli analiti e gli effetti della matrice, e le analisi sono ulteriormente complicate dal fatto che i requisiti variano a seconda del Paese e dei diversi tipi di prodotto. Tra i numerosi prodotti a base di cannabis disponibili in commercio, quelli commestibili rappresentano una delle categorie pi\u00f9 diffuse e comprendono un\u2019ampia variet\u00e0 di prodotti, quali diversi tipi di bevande, cioccolatini, prodotti da forno e caramelle, solo per citarne alcuni. Attualmente lo stato della California richiede l\u2019analisi di una lunga lista di pesticidi e micotossine non solo nel fiore di cannabis, ma anche nei prodotti derivati da questa pianta [1]. Per questa ragione, \u00e8 fondamentale elaborare metodi in grado di affrontare le sfide poste dai diversi tipi di matrice.<\/p>\n\n\n\n<p>Questo articolo descrive lo sviluppo di un metodo di test per l\u2019analisi di pesticidi e micotossine nei brownie alla cannabis in base alla lista stilata dal governo californiano. Abbiamo scelto i brownie come matrice modello in quanto sono molto diffusi tra i consumatori di prodotti alimentari a base di cannabis, ma anche perch\u00e9 contengono livelli elevati di potenziali interferenze (carboidrati e grassi). Illustreremo il lavoro di sviluppo che abbiamo dovuto effettuare per ottimizzare il metodo per l\u2019analisi dei brownie, che proponiamo come punto di riferimento per laboratori che sviluppano metodi per prodotti alimentari simili (per esempio, biscotti o altri prodotti da forno). Il metodo finale qui stabilito per l\u2019analisi di pesticidi e micotossine nei brownie alla cannabis ha fornito risultati eccellenti in termini di linearit\u00e0, accuratezza, precisione e limiti di quantificazione (limit of quantification, LOQ).<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Studio sperimentale<\/h2>\n\n\n\n<p>Inizialmente \u00e8 stato eseguito un lavoro di sviluppo del metodo per ottimizzare la procedura di preparazione dei campioni e per valutare l\u2019impatto degli effetti della matrice sull\u2019analisi quantitativa. L\u2019esito di tali esperimenti viene discusso nella sezione dedicata ai risultati, e viene qui presentata la metodologia finale raccomandata per l\u2019analisi di pesticidi e micotossine nei brownie alla cannabis.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\">Preparazione del campione<\/h3>\n\n\n\n<p>Per un campione solido come i brownie, la prima fase della preparazione del campione \u00e8 l\u2019omogeneizzazione della matrice. Per ridurre il campione in polvere molto fine abbiamo utilizzato un mulino SPEX Freezer\/Mill. I campioni sono stati preraffreddati per 2 minuti, e dopo tre cicli da 2 minuti ciascuno di triturazione (a 15 cps) e 1 minuto di raffreddamento \u00e8 stata ottenuta una polvere sfusa e omogenea facile da lavorare. (In alternativa si potrebbe utilizzare un robot da cucina con ghiaccio secco).<\/p>\n\n\n\n<p>Per il bianco \u00e8 stata pesata una matrice di brownie polverizzata (0,5 g) in un vial di vetro da 4,0 mL (cat.#\u00a024654) e fortificata con pesticidi e micotossine ai livelli definiti nella Tabella I. Una miscela di standard interni \u00e8 stata aggiunta a 200 ng\/g (composti elencati nella Tabella\u202fII). 1,5 mL di acetonitrile acidificato con 1% di acido acetico \u00e8 stato aggiunto al campione. Il campione \u00e8 stato vortexato e sonicato per 5\u202fminuti (non \u00e8 stata necessaria la centrifuga), quindi il surnatante \u00e8 stato fatto passare attraverso una cartuccia SPE Resprep C18 da 100\u202fmg (cat.#\u00a026030). Al pellet del campione sono stati aggiunti ulteriori 1,5 mL di solvente di estrazione (acetonitrile acidificato) e poi il campione \u00e8 stato vortexato di nuovo. Il surnatante \u00e8 stato fatto passare attraverso la stessa cartuccia C18.<\/p>\n\n\n\n<p>Per l\u2019analisi LC-MS\/MS, 750 \u00b5L di surnatante sono stati miscelati a 250 \u00b5L di acqua e il tutto \u00e8 stato poi centrifugato per 5 minuti a bassa temperatura (~7 \u2070C). Un\u2019aliquota di 2 \u03bcL di estratto finale \u00e8 stata iniettata nel sistema LC-MS\/MS.<\/p>\n\n\n\n<p>Per l\u2019analisi GC-MS\/MS, il surnatante rimanente \u00e8 stato trasferito in una provetta Q-sep QuEChERS dSPE contenente solfato di magnesio e PSA pre-pesati (cat.#\u00a026215). Dopo il passaggio al vortex e la centrifuga, 500 \u03bcL di estratti sono stati miscelati a 500 \u03bcL di acetonitrile acidificato. Un\u2019aliquota di 1 \u03bcL di estratto finale \u00e8 stata iniettata nel sistema GC-MS\/MS.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\">Standard di calibrazione e campioni di controllo qualit\u00e0 (QC)<\/h3>\n\n\n\n<p>Una curva di calibrazione a nove punti con un range compreso tra 5 e 700 ng\/g \u00e8 stata preparata in triplicato nella matrice per valutare la linearit\u00e0 e consentire l\u2019analisi quantitativa. Per valutare l\u2019accuratezza e la precisione del metodo, sono stati preparati in quadruplicato tre diversi livelli di concentrazione QC (10, 100, e 500 ng\/g). Sia per gli standard di calibrazione che per i campioni QC, per il bianco sono stati pesati 0,5 g di matrice di brownie polverizzata in un vial di vetro da 4 mL. Nella Tabella I sono riportati i \u00b5L di soluzione madre aggiunti a ogni vial per ottenere concentrazioni diverse nella matrice.<\/p>\n\n\n\n<p><strong>Tabella I<\/strong>: Preparazione dello dello strumento<\/h3>\n\n\n\n<p>L\u2019analisi LC-MS\/MS di pesticidi e micotossine nei brownie alla cannabis \u00e8 stata effettuata utilizzando una colonna analitica Raptor ARC-18 da 2,7\u202f\u00b5m x 100\u202fmm x 2,1 mm (cat.#\u00a09314A12) e uno Shimadzu LCMS-8060 LC-MS\/MS. I parametri generali dello strumento, la polarit\u00e0 ESI di ciascun composto e le transizioni degli analiti sono riportati nella Figura 1.<\/p>\n\n\n\n<p>L\u2019analisi GC-MS\/MS \u00e8 stata effettuata utilizzando una colonna analitica Rxi-5ms (cat.#\u00a013423) da 30\u202fm x 0,25\u202fmm x 0,25 \u00b5m e un Thermo Scientific TSQ 8000 Triple Quadrupole GC-MS\/MS. I parametri dello strumento e le transizioni degli analiti sono riportati nella Figura 2. Si noti che per questa applicazione \u00e8 stato importante utilizzare un liner a cono singolo con lana (cat.#\u00a023447). L\u2019impaccamento in lana aumenta la vaporizzazione e la miscelazione\/omogeneizzazione con il carrier gas per una migliore riproducibilit\u00e0, e il cono nella parte inferiore del liner incanala gli analiti nella colonna, riducendo le potenziali interazioni con la guarnizione dell\u2019iniettore. Inoltre, \u00e8 stato impiegato un tempo di mantenimento di 10 min alla fine di ciascuna corsa per evitare il carry over dei composti e interferenze con le analisi successive.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Risultati e discussione<\/h2>\n\n\n\n<p>Prestazione cromatograficaPer coprire tutti i composti regolati dallo stato di California abbiamo sviluppato i metodi LC-MS\/MS e GC-MS\/MS. Sebbene sia facile analizzare la maggioranza dei pesticidi utilizzando il metodo LC-MS\/MS, i pesticidi idrofobici, quali il chlordane, il methyl parathion, il captan, il chlorfenapyr, e il pentachloronitrobenzene (PCNB), non possono essere rilevati o sono scarsamente ionizzati in condizioni di ionizzazione elettrospray. Altri pesticidi, come il cyfluthrin e la cypermethrin, hanno invece una scarsa risposta in modalit\u00e0 ESI, ma possono essere analizzati sia il metodo strumentale GC-MS\/MS che LC-MS\/MS Al fine di ottenere una risposta ESI accettabile per questi due analiti, le temperature della sorgente di ionizzazione dovrebbero essere impostate su valori relativi bassi. Nel nostro caso, le temperature dell\u2019interfaccia, della linea di desolvatazione e del blocco riscaldante sono state tutte impostate a 100\u202f\u00b0C. In questo studio, chlordane, methyl parathion, captan, chlorfenapyr, and pentachloronitrobenzene (PCNB) sono stati analizzati utilizzando GC-MS\/MS; mentre cyfluthrin e cypermethrin sono stati analizzati utilizzando entrambi gli strumenti, e il resto dei pesticidi e delle micotossine \u00e8 stato analizzato utilizzando solo il metodo LC-MS\/MS. Le Figure 1 e 2 mostrano rispettivamente i cromatogrammi LC e GC per gli analiti target. Uno dei principali vantaggi delle nostre condizioni di gradiente LC \u00e8 che tutte le micotossine eluiscono in 2,4 &#8211; 3,6 minuti, mentre i principali cannabinoidi, quali CBD, CBG, CBN, THC, e THCA-A, cominciano a eluire dopo 5,6 minuti. In base a un confronto con uno standard misto di cannabinoidi analizzati alle stesse condizioni impiegate qui, le uniche coeluizioni tra gli analiti target e i principali cannabinoidi sarebbero tra CBD e piperonyl butoxide (5.7 min), CBG e chlorpyrifos (5.8), e cyfluthrin e CBN (6.5 min). In nessun composto target ci sono state coeluizioni tra THC (delta 8 e 9) e THCA-A.<\/p>\n\n\n<div class=\"wp-block-custom-chromatogram-article-top\"><div A mix of internal standards was added at 200 ng\/g. 1.5 mL of acetonitrile acidified with 1% acetic acid was added to the sample. The sample was vortexed and sonicated for 5 min, then the supernatant was passed through a 100 mg Resprep SPE C18 cartridge (<a target=\"_blank\" href=\"https:\/\/www.restek.com\/p\/26030?utm_source=chromatograms&amp;utm_medium=link&amp;utm_campaign=GC_GN1206\" rel=\"noopener\">cat.# 26030<\/a>). An additional 1.5 mL of extraction solvent (acidified acetonitrile) was added to the sample pellet, and the sample was vortexed again. The supernatant was passed through the same C18 cartridge. After reserving 750 \u03bcL for LC-MS analysis, the remaining supernatant was transferred to a Q-sep QuEChERS dSPE tube containing pre-weighed magnesium sulfate and PSA (<a target=\"_blank\" href=\"https:\/\/www.restek.com\/p\/26215?utm_source=chromatograms&amp;utm_medium=link&amp;utm_campaign=GC_GN1206\" rel=\"noopener\">cat.# 26215<\/a>). Abbiamo utilizzato l&#8217;acetonitrile, il solvente di estrazione utilizzato tradizionalmente nei metodi QuEChERS, perch\u00e9 consente di recuperare una vasta gamma di composti polari e non polari. Inizialmente abbiamo condotto prove sperimentali utilizzando 1 g di campione e almeno 5 mL di acetonitrile per ogni estrazione. Tuttavia, riducendo la dimensione del campione a 0,5 g, siamo riusciti ad abbassare il volume del solvente di estrazione a 3 mL per ciascun campione, pur nel rispetto delle norme californiane. Contenere gli sprechi di solvente rende pi\u00f9 ecologiche le pratiche analitiche adottate e consente di diminuire i costi associati all\u2019analisi del campione e allo smaltimento dei rifiuti. Alla luce di questi vantaggi, per i successivi esperimenti di ottimizzazione sono stati utilizzati campioni da 0,5 g e acetonitrile come solvente di estrazione.<\/p>\n\n\n\n<p>Tra tutti gli analiti target, il daminozide \u00e8 uno dei pi\u00f9 complicati ed \u00e8 stato necessario adeguare ulteriormente la procedura di estrazione per l\u2019analisi di questo pesticida. Il daminozide \u00e8 altamente polare (log P = -1,5), quindi \u00e8 difficile da estrarre completamente dalla matrice di brownie, e non viene trattenuto facilmente in condizioni di fase inversa. Inoltre, i composti a basso peso molecolare, quali il daminozide, sono pi\u00f9 soggetti a interferenze e a un elevato rumore di fondo quando si utilizza MS, soprattutto in caso di scarsa ritenzione cromatografica. Sono state testate diverse strategie per aumentare il recupero di daminozide in condizioni che fossero favorevoli anche per gli altri analiti target.<\/p>\n\n\n\n<p>L\u2019acidificazione del solvente di estrazione \u00e8 stato il primo parametro valutato. A tal fine abbiamo confrontato l\u2019effetto derivante dell\u2019utilizzo dell\u2019acetonitrile con acido acetico (AA) all\u20191% (v\/v) rispetto all\u2019estrazione con acetonitrile puro. La Figura 3 mostra i risultati relativi ai composti target che hanno presentato una differenza di almeno il 40% tra i trattamenti. In caso di estrazione con acetonitrile acidificato, la risposta relativa del daminozide \u00e8 aumentata di circa l\u201980%, mentre la spiroxamine e l\u2019ochratoxin A hanno mostrato rispettivamente un miglioramento della risposta del 45% e 47%. Sulla scorta di quanto emerso e del fatto che nessuno dei composti target ha mostrato una diminuzione significativa della risposta, abbiamo utilizzato acetonitrile con acido acetico all&#8217;1% come solvente di estrazione nel nostro flusso di lavoro finale per la preparazione del campione. Ci\u00f2 offre un ulteriore vantaggio in quanto l\u2019acidificazione degli estratti previene la degradazione del captan, un pesticida adatto all\u2019analisi GC-MS\/MS che si degrada facilmente a valori di pH neutri o elevati [2].<\/p>\n\n\n\n<p>Il secondo parametro per la preparazione del campione che abbiamo esaminato \u00e8 stato l\u2019utilizzo di un\u2019unica estrazione da 3 mL rispetto a un\u2019estrazione in due fasi (da 1,5 mL cadauna). La Figura 4 riassume i risultati relativi agli analiti che hanno mostrato differenze statistiche (test t, valore p &lt;0,05 a un livello di confidenza del 95%) tra le due condizioni di estrazioni (in entrambi i casi \u00e8 stato utilizzato acetonitrile acidificato). \u00c8 interessante notare che il daminozide ha mostrato il miglioramento pi\u00f9 significativo, con un aumento del 40% nella risposta relativa. Gli altri analiti indicati nella figura hanno mostrato un miglioramento della risposta tra il 10 e il 20%. L\u2019aumento delle risposte relative qui riscontrato con l\u2019utilizzo di una seconda fase di estrazione dovrebbe comportare recuperi di estrazione del campione pi\u00f9 elevati in campioni reali. Dato che questo effetto dipende da analita, matrice e solvente di estrazione, si raccomanda di effettuare sempre una valutazione alle condizioni sperimentali desiderate.Dopo l\u2019estrazione con acetonitrile acidificato tutti i campioni sono stati sottoposti a una semplice fase di purificazione con cartucce SPE C18 per rimuovere le interferenze idrofobiche pi\u00f9 significative co-estratte dai brownie. Dopodich\u00e9, i campioni per l\u2019analisi LC-MS\/MS e GC-MS\/MS sono stati trattati in modo diverso. I campioni LC-MS\/MS sono stati diluiti in acqua e centrifugati, rendendo possibile la separazione dei lipidi rimanenti che non erano stati trattenuti nella cartuccia SPE in quanto meno solubili nell\u2019estratto finale una volta aggiunta l\u2019acqua. I campioni GC-MS\/MS sono stati sottoposti a purificazione dSPE con solfato di magnesio e ammina primaria e secondaria (PSA) per rimuovere umidit\u00e0 e zuccheri prima dell\u2019analisi. Questi problemi possono essere superati adeguando il metodo di preparazione del campione applicando condizioni pi\u00f9 selettive, utilizzando uno standard interno idoneo e\/o adeguando il metodo cromatografico per risolvere gli analiti derivanti da interferenze coeluenti. Per questo motivo, dopo aver selezionato le condizioni sperimentali per l\u2019analisi di pesticidi e micotossine nei brownie alla cannabis \u00e8 stata condotta una valutazione degli effetti assoluti della matrice secondo la metodologia proposta da Matuszewski et al. [3]. Per stimare gli effetti assoluti della matrice, sono stati messi a confronto il bianco della matrice e il solvente puro, entrambi fortificati alla medesima concentrazione (15\u202fppb), analizzando la risposta di tutti gli analiti target fortificati. Come illustrato nella Figura 5, solo il daminozide ha mostrato un deciso miglioramento della risposta. Considerando la scarsa ritenzione del daminozide alle condizioni di gradiente LC selezionate per questo studio, \u00e8 probabile che questo pesticida coeluisca con le numerose interferenze non trattenute che sono presenti in genere in matrici come i brownie. Una sintesi dei risultati \u00e8 riportata nella Tabella\u202fIII.<\/p>\n\n\n\n<p>Le curve di calibrazione sono state costruite mettendo a grafico il rapporto tra l\u2019area dell\u2019analita e quella dello standard interno rispetto alle concentrazioni di analiti in standard di matrice di brownie fortificati in un range di 5\u2013700\u202fng\/g. A tutte le curve di calibrazione \u00e8 stato applicato un fattore di ponderazione di 1\/x. La&nbsp;maggioranza dei composti ha mostrato un\u2019eccellente linearit\u00e0, come dimostrato dai valori R2 pari a \u22650,9990 (i valori variavano da 0,9939 a 0,9999).<\/p>\n\n\n\n<p>L\u2019accuratezza e la precisione sono state valutate a tre livelli di concentrazione selezionati per coprire il range lineare: basso (10 ng\/g), medio (100 ng\/g), e alto (500\u202fng\/g). Sono stati ottenuti risultati eccellenti per tutti gli analiti target, perch\u00e9 l\u2019utilizzo di standard preparati in matrice ha ridotto al minimo il potenziamento e la soppressione della matrice. I&nbsp;valori di recupero variavano dal 71,2% al 116% e i valori di RSD erano tutti inferiori al 25%, indicando un buon livello di accuratezza e precisione.<\/p>\n\n\n\n<p>La maggioranza degli analiti ha mostrato valori di LOQ inferiori a 10 ng\/g ed \u00e8 stato dimostrato che la metodologia proposta consente la quantificazione di tutti i composti target a concentrazioni inferiori ai livelli di azione richiesti dallo stato della California per i prodotti a base di cannabis. Il ciflutrin e la cypermethrin hanno mostrato valori di LOQ inferiori in GC-MS\/MS che non in LC-MS\/MS, ma possono comunque essere analizzati ai livelli di azione richiesti con entrambi gli strumenti. Il valore di LOQ per ciascun composto \u00e8 stato definito come la pi\u00f9 bassa concentrazione con un rapporto segnale\/rumore pari ad almeno 10, una differenza inferiore al 25% tra la concentrazione fortificata e la concentrazione stimata, e un valore di precisione delle repliche inferiore al 25%.<\/p>\n\n\n\n<p>La stabilit\u00e0 dei composti target negli estratti LC dopo 24 e 48 ore di stoccaggio nell\u2019autocampionatore a 10\u202f\u2070C (n=4) \u00e8 stata valutata relativamente alle intensit\u00e0 di picco di campioni preparati al momento (0 ore). La maggior parte dei composti ha mostrato un cambiamento nella risposta inferiore al 10% sull\u2019intero periodo oggetto di studio.<\/p>\n\n\n\n<p><strong>Tabella III<\/strong>: Risultati di validazione per il metodo finale ottimizzato per l\u2019analisi di pesticidi e micotossine nei brownie alla cannabis. flusso di lavoro efficace per l&#8217;analisi di pesticidi e micotossine in brownie alla cannabis e sono state illustrate in dettaglio le strategie di ottimizzazione volte a fornire un punto di partenza per matrici simili. Tutti i composti soggetti ad analisi LC-MS\/MS e GC-MS\/MS sono stati estratti impiegando acetonitrile acidificato e successivamente purificati utilizzando cartucce SPE C18. Per i campioni GC-MS\/MS \u00e8 stato necessario effettuare una purificazione dSPE supplementare con solfato di magnesio e PSA. L\u2019utilizzo di acetonitrile acidificato si \u00e8 dimostrato cruciale nel recupero di analiti quali daminozide, spiroxamine e ochratoxin A.
