Gli MDL e gli LLOQ ottenuti con una colonna RMX-5Sil MS sono risultati significativamente pi\u00f9 bassi per circa due terzi dei composti testati rispetto ai risultati ottenuti con una colonna concorrente di fascia premium. \u00c8 stata ottenuta una migliore sensibilit\u00e0 per un\u2019ampia gamma di composti chimici grazie alla deattivazione pi\u00f9 efficace impiegata nelle colonne RMX-5Sil MS.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\"><strong>Introduzione<\/strong><\/h2>\n\n\n\n<p>La quantificazione accurata dei composti organici semivolatili a livello di tracce in matrici ambientali \u00e8 fondamentale per valutare la contaminazione, la conformit\u00e0 normativa e i rischi per la salute umana ed ecologica. Grazie alla sua elevata selettivit\u00e0 e sensibilit\u00e0, la GC-MS\/MS \u00e8 una tecnica di riferimento per abbassare i limiti di rilevazione e quantificazione dei semivolatili. Una maggiore sensibilit\u00e0 consente ai laboratori di adottare metodi di preparazione del campione alternativi, che riducono i volumi di estrazione e, di conseguenza, l\u2019uso di solventi clorurati. La sensibilit\u00e0 del metodo \u00e8 in genere definita da due parametri chiave: il limite di rilevazione del metodo (MDL) e il limite inferiore di quantificazione (LLOQ). L\u2019MDL rappresenta la concentrazione pi\u00f9 bassa che pu\u00f2 essere distinta dai risultati del bianco di metodo con un livello di confidenza del 99%. Al contrario, l\u2019LLOQ definisce la concentrazione pi\u00f9 bassa alla quale un laboratorio<br>dimostra che un analita pu\u00f2 essere quantificato accuratamente.<\/p>\n\n\n\n<p>La riduzione di MDL e LLOQ nei metodi GC-MS\/MS per i semivolatili richiede l\u2019ottimizzazione di molteplici fattori, tra cui la preparazione del campione, le tecniche di iniezione, le condizioni cromatografiche e i parametri dello spettrometro di massa. Inoltre, l\u2019inerzia del percorso di flusso del campione svolge un ruolo critico nella riduzione dei limiti di rilevazione e quantificazione per i semivolatili. I produttori di colonne GC utilizzano diversi processi di deattivazione per neutralizzare la superficie della colonna e prevenire interazioni che riducono la sensibilit\u00e0, causando forme di picco instabili; tuttavia, le deattivazioni tradizionali tendono a funzionare meglio per alcune classi di composti rispetto ad altre. Restek ha sviluppato una deattivazione TriMax di nuova generazione, utilizzata su tutte le colonne RMX, che crea una superficie eccezionalmente inerte ed efficace su un\u2019ampia gamma di classi di composti chimici semivolatili.<\/p>\n\n\n\n<p>Questo studio confronta i limiti di rilevazione e quantificazione ottenibili con una colonna RMX-5Sil MS rispetto alla colonna premium di un altro produttore, valutando i livelli di MDL e LLOQ. Gli esperimenti sono stati condotti utilizzando standard in solvente invece della matrice, cos\u00ec da valutare specificamente le prestazioni della colonna senza l\u2019impatto della manipolazione del campione e dell\u2019estrazione.&nbsp;<\/p>\n\n\n\n<h2 class=\"wp-block-heading\"><strong>Studio Sperimentale<\/strong><\/h2>\n\n\n\n<h3 class=\"wp-block-heading\"><em>Preparazione degli standard e dei campioni<\/em><\/h3>\n\n\n\n<p>Gli standard di calibrazione sono stati preparati in diclorometano a 0,5, 1, 2, 5, 10, 20, 50, 100, 200, 500, 1000, 2000 e 5000 ppb.<br>Per entrambe le colonne, le curve di calibrazione sono state eseguite il giorno 1 insieme a iniezioni in triplicato degli standard 0,5\u2013100 ppb. L\u2019intervallo lineare della curva di calibrazione \u00e8 stato determinato per ciascun semivolatile in base alla risposta del composto.<br>Nei giorni 2 e 3 sono stati preparati standard freschi 0,5\u2013100 ppb, iniettati in triplicato e utilizzati per determinare i valori di MDL e LLOQ per ciascun semivolatile su ciascuna colonna.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\"><em>Condizioni strumentali<\/em><\/h3>\n\n\n\n<p>I campioni sono stati analizzati su una colonna RMX-5Sil MS e una colonna concorrente di fascia premium, entrambe nel formato da 30 m, ID 0,25 mm, 0,25 \u03bcm. L\u2019LLOQ \u00e8 stato determinato sugli stessi campioni ed \u00e8 stato definito come il punto di calibrazione pi\u00f9 basso con un recupero compreso tra l\u201980 e il 120% (oppure la % di recupero disponibile pi\u00f9 prossima) per ciascun semivolatile su ciascuna colonna.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\"><strong>Risultati e Discussion<\/strong>e<\/h2>\n\n\n\n<p>L\u2019MDL \u00e8 risultato pi\u00f9 basso sulla colonna RMX-5Sil MS per il 60% (31\/52) dei composti valutati (Tabella I, Figura 1). Analogamente, l\u2019LLOQ \u00e8 risultato pi\u00f9 basso sulla colonna RMX-5Sil MS per il 63% (33\/52) degli analiti target (Tabella I, Figura 2). I risultati individuali per ciascun composto sono riportati nella Tabella II. Valori di MDL e LLOQ pi\u00f9 bassi indicano che sulla colonna RMX-5Sil MS \u00e8 possibile ottenere una migliore sensibilit\u00e0, attribuibile alla maggiore inerzia garantita dalla nuova tecnologia di deattivazione della superficie di Restek. In particolare, utilizzando la colonna RMX-5Sil MS sono stati ottenuti limiti di rilevazione pi\u00f9 bassi per un\u2019ampia gamma di classi di composti, inclusi semivolatili acidi, basici e massimizzando l\u2019elevata sensibilit\u00e0 ottenibile con strumenti GC-MS\/MS. I risultati presentati in questo studio mostrano che le colonne RMX-5Sil MS, eccezionalmente inerti, offrono una sensibilit\u00e0 maggiore rispetto a una colonna concorrente di fascia premium. Le valutazioni di MDL e LLOQ hanno mostrato che sulla colonna RMX-5Sil MS \u00e8 possibile ottenere limiti di rilevazione e quantificazione pi\u00f9 bassi per un\u2019ampia gamma di composti
